Description
  • CHD subfamily I members CHD1 and CHD2 act as monomeric ATP-dependent chromatin remodellers (reviewed in Alendar and Berns, 2021; Clapier et al, 2017). Human CHD1 and CHD2 are characterized by the presence of double chromodomains at the N-terminal, and central SNF2 helicase ATPase domain, and a C-terminal SANT-SLIDE DNA binding domain (reviewed in Alendar and Berns, 2021; Li et al, 2023). They are 60% identical and 80% similar at the amino acid level (Liu et al, 2015) but have significantly divergent C-terminal regions that may contribute to different functions. Both proteins have been shown to bind to H3K4me3, albeit with different affinities (Sims et al, 2005; Sims et al, 2007; Flanagan et al 2007; Flanagan et al, 2005) and to possess in vitro and in vivo ATP-dependent chromatin remodelling activity (Luijsterberg et al, 2016, Tran et al, 2000; reviewed in Alendar and Berns, 2021; Li et al, 2023). Both proteins are associated with transcriptionally active regions of the genome and may play a role in histone H3.3 deposition (Harada et al, 2012; Konev et al, 2007; reviewed in Alendar and Berns, 2021).
